ACGN Mr. Anime Tournament 2021 (二次元動畫先生專站2021) was the second annual ACGN Mr. Anime Tournament series. It was held after ACGN Miss Anime Tournament 2021 has been ended at the same year. In this tournament, 32 male contestants were eligible to the main tournament. 
Kiyotaka Ayanokouji was the winner for this tournament.
Nomination[]
Nomination Round began on August 1st, 2021 (12:00 a.m.) until August 7th, 2021 (11:59 p.m.)
The rules for nominate characters such as:
- Anime must released in Japan, such as TV Anime, Theatrical anime, OVA and ONA, released before July 20th, 2021. (so "anime" who debuted outside Japan were obviously ineligible).
- Character must be male, and masculine qualities for male with feminime appearances or non-binary (as long as it's not clearly a female). (Characters like
Kurumi Tokisaki was ineligible, with obvious reasons) 
Touma Kamijou, the ACGN Mr. Anime Tournament 2020 champions, was ineligible for this tournaments in the future.
After nomination ended, 32 contestants advanced to the main tournament.
Main Tournament[]
Main Tournament rounds began at August 22th, 2021 until September 17th, 2021. This tournament used 1-vs-1 match method for this round. Unlike the first tournament, this year's tournament used Facebook Like's symbol instead when voting. There were not any playoffs at this year's tournament.
Top 32[]
In this round, 32 contestants divided into 2 "big groups", that contains of 16 contestants at the day of the match. Top 16 contestants with higher scores advanced to the next match and otherwise are eliminated. This match began at August 22nd, 2021, began from 10:00 p.m. (GMT+8), to August 26th, 2021, which lasted for 48 hours.
